Rt Rev Geoff Pearson says the "Bible Challenge" will run in schools in his area from next month until May 2017.
It will encourage children to explore and respond to the Gospel of Luke using new activities every month and rewards.
Using a "Challenge Pack" the children must complete eight activities and for each one completed a sticker will be awarded which will make up part of a bigger picture.
The Diocese of Blackburn believed the initiative is the first of its kind in the Church of England and says other dioceses have already expressed an interest in replicating the format.
All of the activities will be sent to churches and CofE schools but will also be available to parents for free online.
The children who complete all eight activities will be presented with a special Bishop's Bible Challenge Certificate before the end of the summer term in 2017.
Bishop Geoff said: "I want to encourage children to enjoy reading the greatest book in the world - The Bible. It is important for people of all ages to love the word of God if they are to grow in Christian discipleship.
"This fantastic initiative for young people in our church primary schools will help them to engage with The Bible in a brand new way and I am grateful to staff in the Diocesan Board of Education who have helped develop the challenge with me."
