The fire has left a man and a woman dead and five injured at a home in Withington.
Four children aged between ten and 17, and a man, are in hospital. The man is in a serious condition.
An investigation's underway into what caused the fire, which broke out at around 2.30am in Parsonage Road.
Four fire engines tackled the blaze.
Mark Hewerdine, priest in charge at nearby St Chad Ladybarn, said the community have been left in shock.
He told Premier News Hour: "We're quite a close-knit community and many of our members live within just a few doors of where the house fire was, so obviously they've been very shaken and upset by this tragedy. It is particularly moving and particularly upsetting that a family with young children were involved."
He said that the church would be open to anyone in the community who has been affected.
"We will be opening our church from 6 o'clock this evening because people may benefit from a quiet space to think and pray. We will be offering prayers for the family at our services this way and reach out to the family to help them rebuild their lives."
Eye witness Emily Puckering, who lives on the street said: "The first thing I heard was people shouting but the thing I remember is the popping of the windows as the heat made them expand."
